Since chapters 2 and 3 demonstrate that tolmetin and sulindac have antioxidant and free radical scavenging properties, it was decided to determine if these agents could afford protection to rat hippocampal neurons after QA-induced structural damage. Animals were treated with tolmetin or sulindac at a dose of 5mg kg bd for 5 days after which they received unilateral intrahippocampal injections of QA 0.6moles in 2l PBS ; . Histological investigation of neurons in the CA1 and CA3 regions show visible destruction of the structural features of these regions in rats that received QA and significant protection in rats that were pre-treated with either tolmetin or sulindac. QA also induces apoptosis in rat hippocampus but this is attenuated by the treatment of the animals with the NSAIDS. Receptor binding studies were conducted to give an indication of the extent of protection offered by the NSAIDS. In this study, a reduction in the number of NMDA receptors after intrahippocampal administration of QA implied a reduction in the number of viable neurons in the hippocampus. Whilst such a relationship cannot be conclusive in itself, there is a causal relationship between the two. QA significantly p 0.001 ; reduces the number of viable neurons in the hippocampus but the treatment of animals with the NSAIDS reduce the neuronal loss. Tolmetin and sulindac alone had no effect on the number of NMDA binding sites. QA also reduces the viability of primary hippocampal neurons in a concentration-dependent manner. The viability of the neurons was enhanced by the co-incubation of the neurons with either tolmetin or sulindac. The antioxidant and anti-inflammatory properties of these agents maintain the integrity of the neurons. It was postulated that the anti-inflammatory properties of these agents also reduce free radical generation and that the pre-treatment with NSAIDS may have also increased the circulating levels of SOD and toremifene.
