Addition of quinine to the incubation medium induced a sharp drop in the rate coefficient for the efflux of taurine Fig. 7 ; . The maximum inhibitory effect was reached during the subsequent 20 min period. When the quinine-containing medium was replaced by a quinine-free medium, the rate coefficient increased again. During the subsequent 60 min, it reached a peak level which was 83 % higher than the maximum value measured in the cells stimulated by reduced osmolality but not exposed to quinine. During the remaining part of the incubation period, the rate coefficient declined towards the level found in the cells not treated with quinine. Bumetanide, DIDS, furosemide and quinine inhibited both the Na + -independent, osmolality-stimulated influx of taurine and the volume regulatory efflux of taurine Fig. 6; S. M. Thoroed, M. S. Soegaard, E. J. Cragoe and K. Fugelli, unpublished results ; . An interesting question arising from these results was whether the two fluxes could be mediated by the same transport system. The relationship between the rate coefficient k ; for a transport process, the flux J ; and the concentration of the substance transported [S] on the cis side of the membrane is given by the equation: k J [S]. The influx measurements Fig. 5 ; were carried out at a constant extracellular concentration of taurine. Thus, the relative effects of the various inhibitors on the rate coefficient for the volume regulatory efflux of taurine were comparable with the corresponding effects on the Na + -independent influx. This comparison is shown in Fig. 8. A strong correlation was revealed.

The train-whistle sounds and the train from the Harlem division of the New York Central slowly pulls into the quiet station in the tiny hamlet. A large boxcar opens up and three huge elephants are guided off the car past the freight house. The elephants don't need to be prodded. They are very familiar with the station. They have just returned from their last show at the Palace Theatre, and they laboriously begin the slow walk home along the dirt road that leads to their barn.

Evidence of additive antiviral activity and the absence of additive cytotoxicity of TG and AZT to host lymphocytes indicates that TG warrants further evaluation as a component of antiviral chemotherapy. These experiments establish that incorporation of dGSTP affects both steps of viral replication--DNA polymerization and RNA cleavage; therefore, thiopurines represent a new class of agents with anti-retroviral activity. The currently available non-nucleosides inhibitors of HIV-1 RT interfere with RNase H 34 ; and polymerase 35 ; functions by binding a specific region of the p66 subunit of HIV-1 RT in a hydrophobic pocket of the palm domain, and mutations in this binding site have been shown to confer HIV-1 resistance to these agents. Because thiopurines inhibit RNaseH function by modifying its DNA-RNA substrate, and not by directly binding HIV RT, its anti-HIV activity should not be impaired by these HIV-RT mutations. Thus, the present work establishes that thiopurine medications, which have been in clinical use for almost 50 years, possess novel anti-retroviral effects that may complement contemporary agents used to treat HIV infected patients.

By the time the diagnostic criteria for MH was met and treatment initiated, the rigidity was so severe that pulses could no longer be detected in the extremities, despite adequate central arterial pressures. It is unlikely that the skeletal muscle could achieve therapeutic dantrolene concentrations with perfusion compromised to this extent. Since the MH episode did not resolve despite multiple doses of both MC-NaD and D, treatment failure was thought to be the result of initiating treatment too late in the course of the episode.
