BACKGROUND: Azapyrimidine nucleosides are an interesting class of nucleosides which are derived from the replacement of the carbon atom of a pyrimidine ring by nitrogen. For example, D-6-azacytosine is an antimetabolite with wide spectrum of therapeutic activities. D-6-Azauridine has exhibited antitumor activity both experimentally and clinically. METHODS: The synthesis of carbocyclic L-6-azapyrimidine nucleosides was based on the regiospecific coupling of an enantiomerically pure L-cyclopentanol pseudo sugar moiety with an appropriate protected 6-azapyrimidine base. Starting from D-ribose, a new methodology for the synthesis of L-4, was developed via a ring-closing metathesis to prepare L-cyclopentanol. By choosing 5-methylthiotriazinone as N3-blocked base, we realized the N1-regiospecific coupling of 6-azapyrimidine base with L-cyclopentanol under standard Mitsunobu conditions. Preliminary antiviral activities against respiratory virus of the synthesized nucleosides are evaluated. RESULTS: Three carbocyclic L-6-azapyrimidine nucleosides were synthesized and evaluated for their antiviral activities against SARS, yellow fever, West Nile Virus, VEE, and Punta Toro A virus, however no significant antiviral activity against these virus was detected.
